Cell Details
The synovial membrane, which is the inner layer of the joint capsule. It is light red, smooth and shiny, thin and soft, and consists of loose connective tissue. All structures in the joint cavity, except the articular cartilage and the meniscus cartilage plate, are even covered by the synovium through the tendons and ligaments of the joint cavity.
The synovial membrane secretes synovial fluid and plays an important role in joint activity. The normal synovium is divided into two layers, namely a thin cell layer (inner lumen layer) and a vascular layer (intimal sub-layer), which is a blood vessel-rich joint capsule intima, attached to the non-articular surface and covered in the joint capsule. On the bone surface, not on the cartilage surface, this part is called the marginal zone or the "naked zone". The synovial membrane is pink, smooth, shiny, wet and lubricated, sometimes with fluff and containing collagen fibers.
There are two types of synovial cells: A and B. Macrophage-like A-type cells with filamentous pseudopods, serosal invagination, vesicles, mitochondria, lysosomes, cytoplasmic fibers, and Golgi, with phagocytic function; B-type fibroblast-like synoviocytes (FLS) ), with a high concentration of endoplasmic reticulum structure, is the main cell that mediates the destruction of RA joints.
Cell Characteristics
1) The cells are derived from surgery to obtain normal synovial tissue of the human joint.
2) Cell identification: Fibronectin or vimentin was positive for immunofluorescence staining.
3) The purity of the identified cells is higher than 90%.
4) Does not contain HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi.
5) Cell growth mode: fibroblast-like cells, adherent culture.
Transportation and Preservation
Depending on the weather conditions and the distance of transportation, the company negotiates with the customer and chooses one of the following methods.
1) 1mL of frozen cell suspension is placed in a 1.8mL cryotube and placed in a foam incubator filled with dry ice for transport; after receiving the cells, thaw the resuscitated cells as soon as possible for culture. If resuscitation is not possible immediately, Cryopreserved cells can be stored at -80°C for 1 month.
2) T-25 culture flask is filled with complete medium and then transported at room temperature. After receiving the cells, please observe the growth state of the cells under a microscope. If the bottle filling rate exceeds 85%, please carry out the subculture immediately. If there are more cells in suspension, allow the flask to stand overnight in the incubator to help the undead suspension cells to reattach.
